Ein ausserst ernsthaftes Problem der landwirtschaftlichen Produktion stellt die Kontamination von Futtermitteln mit Mykotoxinen dar. Dabei kann die Gesundheit und vor allem die Leistungsfahigkeit der Nutztiere beeintrachtig sein. Daruber hinaus besteht ebenfalls eine Gefahrdung des Menschen durch Aufnahme von mykotixinhaltigen Lebensmitteln tierischer oder pflanzlicher Herkunft. Mykotoxine gewannen in den letzten 20 Jahren standig an Bedeutung als Ursache fur Fortpflanzungsstorungen im Nutztierbestand. Dabei spielen die von Fusarienarten gebildeten Toxine Zearalenon (ZON) und Deoxynivalenol (DON) die grosste Rolle. Eine starke Anreicherung und Verbreitung dieser Feldpilze ist die Folge ackerbaulicher Massnahmen. So wird aus Zeit- und Kostenersparnis eine pfluglose Bodenbearbeitung bevorzugt. Die fehlende Tiefe dieser Bodenbearbeitungsmethode fordert das Pilzwachstum enorm. Weiterhin wird die Entwicklung der Pilze besonders von den klimatischen Bedingungen beeinflusst. Feuchte und warme Sommer sind ideale Voraussetzungen fur das Wachstum von Fusarien. Eine besonders ausgepragte Empfindlichkeit gegenuber Zearalenon zeigen Schweine. Dabei sind die klinischen Erscheinungen abhangig vom Geschlecht und dem Alter der Tiere. Dieses Buch beschaftigt sich mit der Bestimmung von Zearalenon sowie seiner Metabolite a-Zearalenol und ss-Zearalenol im Gallensaft von Schweinen mittels LC/MS-MS. Ziel der vorliegenden Studie war es, eine bereits bestehende Standardmethode zur Probenaufarbeitung von Gallensaft durch Variation bei der Festphasenextraktion zu optimieren. Die dabei untersuchten SPE-Materialien und Methoden wurden hinsichtlich ihrer Effizienz, ihres Zeitbedarfs sowie ihrer Kosten verglichen und bewertet. Weiterhin wurde eine herkommliche Detektionsmethode DAD durch die Massenspektrometrie ersetzt.
